Antagonism of the MCP-1 receptor (CCR2) slowed C5a mediated HSC migration. HSC on day 8 of culture were used in a two-dimensional wound healing assay (A,B). Cells were pre-treated with or without a CCR2 receptor antagonist, 227016 (10 nM), for 30 min and then challenged with C5a (10 ng/mL) and/or PDGF (50 ng/mL). Images were captured at 0 h, 10 h, and 24 h following stimulation. Gap width was analyzed by Wimasis image analysis software. Values represent means ± SEM, N = 4.
