Images were captured at 0 h, 10 h, and 24 h. Gap width and rate of wound healing were analyzed with Wimasis image analysis software. (C) Boyden chamber chemotaxis: On day 8 of culture, HSC were serum starved for 18 h, then detached and resuspended in serum-free DMEM. Inserts containing cells were immersed in serum-free DMEM with or without 10 ng/mL C5a, 50 ng/mL PDGF or both. Chemotaxis was assessed after 10 h incubation, as described in Methods.
